Right whats the best way to set the mixture on the carbs, is the colourtune any good at all or another american ripoff, or do people go for the pressing the plunger methord and listening to the engine revs.

Personally, I used the plunger to get the basic setup because it is fairly quick, but it left it a bit rich, I used Colortune to refine it and based my final setting on the plug colour over a period of time.

Colortune works very well. Sounds like you have twins from your threads though, so using two Colortunes is the best option, one in #1 and one in #4. Also use a Carbalancer if you have twins. Tuning to the right point of the mixture plateau is the proper SU tuning method but is fiddly and tricky especially with twins, Colortune works well. You can't do full load tuning using this though so it only really works once you have the right needles, brilliant on a standard car or after a rolling road has set the needles up properly but initially on a new custom build an RR session is still needed.

does the minispares colourtune kit come for the mini only (large spark plug hole) or does it have an adapter for a smaller spark plug hole too?

Watch that stuff. Colortune is made by Gunson... a British firm... NOT American.
